Key points for using polyethylene glycol
¡¡Hits£º65¡¡AddTime£º2026/2/28 10:36:49
Polyethylene glycol (PEG) is a polymer material widely used in various fields, from chemical engineering to pharmaceuticals, from daily chemical products to biotechnology. Due to its wide range of molecular weights (from liquids to solids) and vastly different application scenarios, the key points of use need to be differentiated based on specific applications.

In order to help you quickly grasp the core points, I have compiled an overview table of key considerations in different scenarios, and will provide detailed explanations for several main areas later.

📋  Overview of key points for using polyethylene glycol
Common molecular weight choices in application fields ⚠️  Key usage points
Medical PEG 4000 (constipation), PEG 3350/4000 (intestinal preparation), and PEG (eye drops) are strictly distinguished for oral, topical, and instrumental use; Be alert to the risk of intestinal obstruction when taking orally; Pay attention to the expiration date (usually 1 month) and bottle mouth contamination prevention of eye drops after opening the lid.
In vitro diagnostic (IVD) PEG 6000/8000 (immunoturbidimetry), PEG 20000 (colloidal gold), PEG 400-4000 (enzyme reaction) concentrations are the soul and require precise optimization, as excessive concentrations can lead to protein denaturation; Specific molecular weights must be selected based on the application scenario and cannot be mixed; Select high-purity diagnostic grade to avoid interference from impurities in the detection results.
Chemical/industrial PEG 200-600 (liquid), PEG 20000 (solid). Pay attention to their flammability and dust explosion risks, and stay away from sources of fire and heat; Pay attention to personal protection (gloves, goggles) during operation, and avoid long-term or repeated skin contact; Waste disposal must comply with local environmental regulations.
Biological/pharmaceutical research and development of specific length PEGs (such as PEG4, PEG12) for PEGylation modification as drug excipients can increase the solubility of insoluble drugs, improve drug stability, and reduce drug irritation; In protein modification, monodisperse and functionalized PEG derivatives (such as NHS esters and maleimide) should be selected to achieve site directed coupling.
💊  Medical use: Distinguishing the route of administration is the top priority
In the medical field, PEG is most commonly used as a laxative (such as polyethylene glycol 4000 powder) and intestinal cleanser (such as polyethylene glycol electrolyte powder) for oral administration, and also as an eye drop to alleviate dry eye syndrome.

Oral bowel preparation: When preparing for colonoscopy with polyethylene glycol electrolyte powder, it is usually necessary to drink a large amount of water (about 2-4 liters) and take it at a certain speed. If there is severe abdominal distension or pain, the medication should be paused or slowed down. It is forbidden for patients with intestinal obstruction, gastric retention and gastrointestinal bleeding.

External eye drops: Polyethylene glycol eye drops usually have a usage period of one month after opening. Do not use after expiration. When dripping, do not let the bottle mouth touch your eyes or hands to prevent contamination of the medication.

🔬  In vitro diagnosis: precise game between concentration and molecular weight
In diagnostic reagents, PEG plays multiple roles such as signal enhancer and stabilizer. Whether it can be used well directly determines the sensitivity and stability of the detection reagent.

Concentration is key: PEG concentration is too low to be effective, while high concentration (such as>20%) may lead to the aggregation and denaturation of antibodies and other proteins, which can interfere with detection.

The molecular weight should match:

Immunoturbidity: PEG 6000 or 8000 is commonly used to accelerate the formation of antigen antibody complexes through its "exclusion effect".

Colloidal gold chromatography: High molecular weight PEG 20000 is commonly used to stabilize gold conjugate particles, prevent their aggregation, and adjust the chromatography rate.

Enzyme reaction system: Low molecular weight PEG 400-4000 is commonly used to protect the activity of tool enzymes.

🧪  Industry and R&D: Balancing Safety and Functionality
In chemical production and laboratory research and development, the focus may be completely different when dealing with barrels of industrial raw materials or high-value specialized reagents.

Industrial safety: Solid PEG (such as PEG 20000) is a combustible material, and its dust may form explosive mixtures when mixed with air. Storage should be cool, ventilated, and separated from oxidants. Dust masks and chemical protective goggles should be worn during operation.

Protein modification (PEGylation): This is a cutting-edge technology in biopharmaceuticals. At this time, not ordinary PEG is used, but monodisperse PEG with specific active groups at one or both ends (such as NHS ester, maleimide). They are like precise "LEGO bricks" used to connect proteins and extend the half-life of drugs.
